The victory at Bramall Lane pushed Sheffield United closer to relegation, as they are now 10 points behind safety with only five games left.
Goals from Jacob Bruun Larsen and Lorenz Assignon in the first half gave Burnley a solid lead by halftime.
Although Gustavo Hamer briefly gave Sheffield United hope with a goal, Burnley secured the win with goals from Lyle Foster and Johann Berg Gudmundsson.
It was a good day for Burnley as their relegation rival Luton suffered a heavy defeat at home against Brentford, losing 5-1.
Luton, currently in 18th place, had the opportunity to climb out of the bottom three with a win, but they are now just two points ahead of Burnley in 19th place.
Yoane Wissa scored twice for Brentford, who now have a comfortable 10-point lead over the relegation zone.
